Evaluación del efecto de la proteina GP63 sobre linfocitos B humanos (Ramos RA-1) y células mononucleares de sangre periférica

| dc.description.abstract | La proteína GP63 es una metaloproteasa presente en la membrana de parásitos del género Leishmania spp., que le permite evadir la respuesta inmune del hospedero. Esta proteína modula diversas moléculas del sistema inmune, incluyendo el C3b del complemento, el correceptor CD4 en linfocitos T, mTOR, caspasas, algunas MAPK quinasas y factores de transcripción como NFκB. Su capacidad para modular múltiples rutas inmunológicas fue lo que permitió considerarla una posible herramienta para modular blancos inmunológicos relevantes en linfocitos B (LB). Inspirados por los avances recientes en el desarrollo de proteínas recombinantes y la transferencia dirigida de proteínas o material genético con fines terapéuticos; se desarrolló un sistema de expresión constitutiva de la proteína GP63 y la producción de la proteína recombinante. Estos modelos, permitieron evaluar los efectos de GP63 en la supervivencia, proliferación, activación y diferenciación de LB humanos. Observamos que la exposición a GP63 recombinante, inducía la expresión de CD38, mientras que su expresión intracelular generaba el aumento de CD11b. En conjunto, estos cambios sugieren que la vía de dosificación de la proteína tiene un impacto en el fenotipo del LB, en especial hacia un perfil más regulador que activador. Mediante esta aproximación biotecnológica, buscamos aportar evidencia crítica sobre la aplicación de este tipo de proteínas como agentes terapéuticos. El enfoque de este trabajo es innovador, ya que se aleja del paradigma tradicional que considera a GP63 exclusivamente como un factor de virulencia o un blanco terapéutico, proponiendo en cambio su aprovechamiento como una molécula inmunomoduladora, con potencial bioactivo, en contextos terapéuticos que puedan ser aplicados en el futuro, para el tratamiento de enfermedades inmunomediadas. | spa |
| dc.description.abstract | The GP63 protein is a metalloprotease present in the membrane of parasites of the genus Leishmania spp., which enables them to evade the host’s immune response. This protein modulates various molecules of the immune system, including complement component C3b, the CD4 receptor on T lymphocytes, mTOR, caspases, certain MAPK kinases, and transcription factors such as NFκB. Its ability to modulate multiple immunological pathways was the key feature that motivated its study in this thesis, considering it as a potential tool to modulate relevant immunological targets in B lymphocytes (BL). Inspired by recent advances in the development of recombinant proteins and the targeted delivery of proteins or genetic material for therapeutic purposes, we developed a constitutive expression system for the GP63 protein and produced the recombinant protein. These models allowed us to evaluate the effects of GP63 on the survival, proliferation, activation, and differentiation of human B lymphocytes. We observed that exogenous exposure to GP63 induced CD38 expression, whereas its intracellular expression led to an increase in CD11b. Taken together, these changes suggest that the protein delivery route influences the B cell phenotype, particularly shifting it towards a more regulatory rather than an activating profile. Through this biotechnological approach, we aim to provide critical evidence for the application of such proteins as therapeutic agents. This work takes an innovative approach by moving away from the traditional paradigm that considers GP63 solely as a virulence factor or therapeutic target and instead proposes its use as an immunomodulatory molecule with potential bioactivity in therapeutic contexts that could be applied in the future for the treatment of immune-mediated diseases. | eng |
